Go to Col'Cacchio and order as many types of pizza as you can between you and your friends. You won't regret it.
Go to De Akker on Dorp Street if you're looking for a pub-style bar. It's the oldest bar in Stellenbosch, and it attracts an older crowd than the usual university group in town. For a good game of pool and good music every once in a while, check out Stones.
Check out either Bohemia or Die Mystic Boer. You might not understand the language in which the bands are singing, but both places have live music regularly.
Watch res rugby at the University fields. It's a ton of games and a ton of people all on one field. For an American football lover, it's cool to see other die hard fans for sports that cause concussions. Also, seeing the way these guys hit up close (with no pads, or cups) will make you rethink any previous notions of the word "toughness." Or stupidity? Just don't mention Austrailia if you want to make friends.
Watch your back. Not an incredibly dodgy place by any means, but ladies, don't walk home by yourself at night.
Know that a lot of people in Stellenbosch speak Afrikaans, and they'll probably speak it to you out of habit. Just say this: Ek prat nee afrikaans nee. Not how you spell it at all, but if I spelled it for you there'd be no hope of pronouncing it.
